British star Skupski and Dutchman Koolhof have established themselves as the best pair in the world over the past 18 months but are yet to win a grand slam title together.
The top seeds found themselves on the back foot early on and were unable to force a break point throughout the match against Spaniard Granollers and Argentina's Zeballos.
Skupski and Koolhof took a 3-1 lead in the tie-break as they tried to force a deciding set but lost six of the next seven points in a 6-3 7-6 (4) defeat.
