Kuznetsova has been struggling with a knee injury and on Tuesday confirmed that she would not be fit to take her place at Flushing Meadows.
Others to pull out of the women’s draw are world number fifteen Kaia Kanepi (Achilles) and Flavia Pennetta (wrist). They are joined on the sidelines by 2010 finalist Vera Zvonareva who has been suffering with a virus since Wimbledon.
Briton Elena Baltacha has already announced that ankle surgery will keep her from the competition.
Kuznetsova, Kanepi, Pennetta and Baltacha will be replaced by Greta Arn, Johanna Larsson, Garbine Muguruza and Sesil Karatantcheva. A replacement for Zvonareva has yet to be named.
On the men’s side of the draw, Juan Carlos Ferrero - a runner-up in 2003 - has pulled out.
Ferrero joins compatriot and world number three Rafael Nadal, Gael Monfils and Juan Ignacio Chela on the sidelines.
